Ampliqon Taq DNA Polymerase is a recombinant thermostable DNA polymerase originating from the thermophilic bacterium Thermus aquaticus.

Ampliqon Taq DNA polymerase is available in different standalone variants, concentrations and master mixes.

Standalone variants are supplied either without buffers or in kits including MgCl2 and one or two Ampliqon PCR buffers to achieve highest PCR performance and to avoid tedious reaction optimisation.

Master mixes include all components required for high performance PCR, just add your specific primers and DNA template.

Taq DNA Polymerase

Ampliqon Taq DNA Polymerase 5 U/µl  is ideal for routine PCR applications requiring high yield and also reliable DNA amplification. Taq DNA Polymerase 5 U/µl  is available in different sizes.

DESCRIPTION

This has a molecular weight of 95 kDa and exhibits both a 5’→3′ DNA polymerase and a 5’→3 exonuclease activity. The 5′-3′ exonuclease activity  leaves a 3’dA overhang on the PCR product, which are convenient for direct T-A cloning. Taq DNA Polymerase lacks 3′-5′ exonuclease activity and proofreading ability.

Taq DNA polymerase is suitable for standard testing, routine PCR, screening and high throughput testing.

Ampliqon Taq DNA Polymerase was compared with a Taq DNA Polymerase from a well-recognised competitor. Three different DNA target varying in length PAH (203 bp), ENG9 (293 bp) and ENG 5 (480 bp) were evaluated. Ampliqon Taq DNA Polymerase performed equally well or better in this study. Furthermore, Ampliqon Taq DNA Polymerase displays a higher specificity on the PAH and ENG5 targets, than the competing Taq DNA Polymerase. Each Amplification has been conducted according to supplier’s manual.

Taq DNA Polymerase RED

Taq DNA Polymerase RED 5 U/µl is suitable for routine PCR applications requiring high yield and also reliable DNA amplification. Taq DNA Polymerase RED is a good alternative to Ampliqon Taq DNA Polymerase. The red dye in this product provides convenient identification of enzyme addition to the tube and also confirms complete mixing. Taq DNA Polymerase RED is available in different sizes.

DESCRIPTION

The inert red dye has no effect on downstream processes but adds visibility to the enzyme. This makes Taq DNA Polymerase RED very useful for high throughput testing. Taq DNA Polymerase RED is added directly to the PCR reaction mix.

Taq OptiMix CLEAR is an optimised format of Taq DNA Polymerase Master Mix and is therefore a good alternative to Taq DNA Polymerase and Taq DNA Polymerase Master Mix.  Taq OptiMix CLEAR ensures the user an increased specificity and improved PCR performance, even on difficult templates. Taq OptiMix CLEAR offers reduced set-up time and eliminates the risk of contamination of reaction components. Taq OptiMix CLEAR is suitable for standard PCR applications, screening and high throughput testing.

COMPARISON BETWEEN TAQ OPTIMIX CLEAR AND THREE COMPETITORS

Taq OptiMix CLEAR was compared with corresponding Taq DNA Polymerase master mixes from three well recognised competitors; T,K and N. Six different primer targets (BAIP, Q8, CFTR, ENG5, ENG9 and PAH) were evaluated. Taq OptMix CLEAR performed equally well or better on all six primer targets tested in this set up. Each amplification was conducted according to supplier’s manual.

This is a very popular alternative to Taq DNA Polymerase Master Mix with the same advantages. The inert red dye and stabiliser are extra features, which allow direct loading onto your agarose gel for analysis.

DESCRIPTION

The Master Mix RED is a ready to use 1.1x or 2x master mix. Taq DNA Polymerase Master Mix RED is available in two final MgClconcentrations: 1.5 mM and 2.0 mM. Taq DNA Polymerase Master Mix RED is composed of Ampliqon Taq DNA Polymerase, the NH4+ buffer system, dNTPs and magnesium chloride.

The red dye and the stabiliser do not interfere with the PCR. If necessary, the red dye can be removed from the PCR product by spin column purification or other methods. Furthermore, PCR products generated using Taq DNA Polymerase Master Mix RED DNA, can directly be used for sequencing after treatment with either spin columns or PureIT ExoZAP PCR CleanUp. 

DIRECT GEL LOADING

After end of amplification PCR products can be loaded directly onto agarose and SDS DNA gels. The red dye provides easy visualisation of both pipetting and gel loading. The red dye front runs at 300 – 1000 bp on 0.5 – 1.5 % agarose gels.

RED DYE EXHIBITS NO PCR INTERFERENCE

The red dye does not interfere with PCR performance. Two different DNA targets; PAH (203 bp) and BAIP (788 bp) were amplified in duplicates using Taq Master Mix RED and visualised on a agarose gel.
