Listeria monocytogenes, described in 1926 by Murray, Webb, and Swann, is a widespread problem in food industries. Evidence from outbreaks of listeriosis indicate the principle route of transmission is via the consumption of foodstuffs contaminated with Listeria monocytogenes. Implicated vehicles of transmission include turkey frankfurters, coleslaw, pasteurized milk, Mexican style cheese, and pate. PALCAM Broth is based on the formulation of Van Netten et al. while he was investigating the isolation of Listeria spp. from food samples. PALCAM Broth has been used for testing Listeria from raw milk.

GC Agar is used with lysed blood and other supplements for the isolation and cultivation of Neisseria gonorrhoeae and other fastidious organisms. GC Agar is not intended for use in the diagnosis of disease or other conditions in humans.
A nutritious agar base described by Thayer and Martin for the isolation of Neisseria gonorrhoeae. The rich peptone mixture is enhanced by the use of corn starch to absorb toxic metabolites and a buffering system is used to maintain neutral pH. The medium is made selective by the use of various antibiotic cocktails. Thayer and Martin originally recommended the use of vancomycin, colistin and nystatin and the addition of trimethoprim which is useful in preventing the swarming of proteus. More recently the emergence of vancomycin sensitive gonococci has made the selective agents (lincomycin, colistin, amphotericin, trimethoprim (X070 LCAT) the combination of choice. Enrichment of the base is usually by the addition of lysed blood. Alternatively, chocolated blood or hemoglobin powder and Thayer and Martin’s mixture of vitamins, amino acids and coenzymes can be used. GC Growth Supplement (X271) can be added to this medium to aid in the isolation of Neisseria spp.

Yeast Extract is an autolysate of yeast cells used in preparing microbiological culture media in a laboratory setting. Yeast Extract is not intended for use in the diagnosis of disease or other conditions in humans. Yeast Extract is the water-soluble portion of autolyzed yeast. The autolysis is carefully controlled to preserve naturally occurring B-complex vitamins. Yeast Extract is prepared and standardized for bacteriological use and cell cultures and is an excellent stimulator of bacterial growth. Yeast Extract is generally employed in the concentration of 0.3% – 0.5%. Yeast Extract is typically prepared by growing baker’s yeast, Saccharomyces spp., in a carbohydrate-rich plant medium. The yeast is harvested, washed, and resuspended in water, where it undergoes autolysis, i.e., self-digestion using the yeast’s enzymes. Yeast Extract is the total soluble portion of this autolytic action. The autolytic activity is stopped by a heating step. The resulting Yeast Extract is filtered clear and dried into a powder by spray drying. Yeast Extract has been successful in culture media for bacterial studies in milk and other dairy products. Several media containing Yeast Extract have been recommended for cell culture applications.

Tryptic Soy Broth (Soybean-Casein Digest Broth) is used for the cultivation of a wide variety of microorganisms. Tryptic Soy Broth conforms to Harmonized USP/EP/JP requirements and is not intended for use in the diagnosis of disease or other conditions in humans. Tryptic Soy Broth, a general purpose medium, is commonly referred to as Casein Soya Bean or Soybean-Casein Digest broth, and abbreviated as TSB. This medium was originally developed for use without blood in determining the effectiveness of sulfonamides against pneumococci and other organisms. Clostridia and non-sporulating anaerobes grow luxuriantly in this broth when incubated under anaerobic conditions. TSB is recommended for testing bacterial contaminants in cosmetics and complies with established standards in the food industry. TSB was chosen by the USDA Animal and Plant Health Inspection Service for detecting viable bacteria in live vaccines. TSB conforms to Harmonized United States Pharmacopoeia (USP), European Pharmacopoeia (EU), and Japanese Pharmacopoeia (JP). The rich nutritional base of TSB, supplemented with SPS and CO2, is an excellent broth for blood cultures. With the addition of 6.5% NaCl, TSB can be used for the selective growth of group D streptococci. Enzymatic Digest of Casein and Enzymatic Digest of Soybean Meal are nitrogen sources in TSB. Dextrose is the carbon energy source that facilitates organism growth. Sodium Chloride maintains osmotic balance; Dipotassium Phosphate is a buffering agent.

In 1948, Weber and Black described the value of a highly nutritional solid medium containing neutralizing agents for quaternary ammonium compounds in sanitizers. The addition of Lecithin and Polysorbate 80 to Tryptone Glucose Extract (TGE) Agar resulted in a medium that effectively neutralizes quaternary ammonium compounds while testing germicidal activity. Letheen Agar is a modification of TGE Agar with the addition of Lecithin and Polysorbate 80.
Letheen Broth Base was developed as a subculture medium for the neutralization of quaternary ammonium compounds in disinfectant testing. Quisno, Gibby, and Foter discovered that adding Lecithin and Polysorbate 80 to F.D.A. Broth resulted in a medium that neutralized high concentrations of quaternary ammonium salts. The resulting medium, termed “Letheen” (a combination of Lecithin and Tween) was easy to prepare and clear in appearance, aiding to visual inspection for growth. Letheen Broth Base is recommended by the Official Methods of Analysis of the Association of Official Analytical Chemists (AOAC) for use with disinfectants containing cationic surface active materials.

In 1955, Leavitt et al. discovered Tryptic Soy Agar supported excellent growth of aerobic and anaerobic microorganisms. Tryptic Soy Agar is a nutritious base and a variety of supplements are added to enhance the medium, including Lecithin and Tween 80. The Lecithin and Tween 80 inactivate some preservatives that may inhibit bacterial growth, reducing “preservative carryover”. Tryptic Soy Agar with Lecithin and Tween 80 is recommended for determining the sanitation efficiency of containers, equipment, and work area (environmental monitoring).

LB Agar (Miller), a nutritionally rich medium designed for growth of pure cultures of recombinant strains, is based on the formulation described by Miller. E. coli is grown to late log phase in LB Medium. Some plasmid vectors replicate to a high copy number and do not require selective amplification. Some vectors do not replicate so freely and need to be selectively amplified. Chloramphenicol may be added to inhibit host synthesis and prevent replication of the bacterial chromosome.
LB Agar, Miller contains 10 g/L of sodium chloride, different from the levels in Lennox formulations. This allows the researcher to select the optimal salt concentration for a specific strain. The medium may be aseptically supplemented with glucose or antibiotics.

Christensen devised a urea agar medium containing peptone and dextrose that had a reduced buffer content. The medium supported the vigorous growth of many Gram-negative, enteric bacilli and readily permitted observation of urease production. Urea Agar Base may be used as a screening medium (along with Triple Sugar Iron Agar) for the selection of Salmonella and Shigella cultures for serologic classification. Urea Agar Base is also used to detect production of urease by yeast. Urease production is an important differential test in microbiology and outlined in standard methods.

Simmons Citrate Agar is used for the differentiation of microorganisms on the basis of citrate utilization in a laboratory setting. Simmons Citrate Agar is not intended for use in the diagnosis of disease or other conditions in humans.
Koser first developed a liquid medium for differentiating coliforms from fecal coliforms. Fecal coliforms were unable to use citrate as the sole source of carbon and inorganic ammonium salt as a sole source of nitrogen. Non-fecal coliforms, such as Enterobacter aerogenes could use citrate in such a medium with resultant alkalinity. Liquid medium had the disadvantage of appearing turbid when a large inoculum was used, although no growth had taken place. This observation led Simmons to devise a solid medium that eliminated the problem with turbidity.
Simmons Citrate Agar is a modification of Koser’s medium, with the addition of bromothymol blue and 1.5% agar. Organisms able to metabolize citrate grow luxuriantly. The medium is alkalinized and changes from green to deep blue in 24 – 48 hours. Escherichia coli either do not grow at all on this medium or grow so sparsely that no change in reaction is apparent. Simmons Citrate Agar is recommended for differentiation of enteric Gram-negative bacilli from laboratory specimens, water samples, and food samples.

D/E Neutralizing Broth is used to neutralize and determine the bactericidal activity of antiseptics and disinfectants. D/E Neutralizing Broth without Tween is not intended for use in the diagnosis of disease or other conditions in humans. D/E Neutralizing Broth was developed by Dey and Engley to neutralize a broad spectrum of disinfectants and preservative antimicrobial chemicals, including quaternary ammonium compounds, phenolics, iodine, chlorine preparations, mercurials, formaldehyde, and glutaraldehyde. D/E Neutralizing media neutralize higher concentrations of residual antimicrobials when compared with other standard neutralizing formulations, such as Letheen media, Thioglycollate media, and Neutralizing Buffer. Total neutralization of disinfectants is critical. Disinfectant residues can result in a false-negative (no-growth) test. D/E Neutralizing Broth without Tween effectively neutralizes the inhibitor action of disinfectant carryover, allowing differentiation between bacteriostasis and the true bactericidal action of disinfectant chemicals. This is a critical characteristic to consider when evaluating a disinfectant. D/E Neutralizing Broth without Tween is recommended for use in disinfectant evaluations, environmental sampling (swab and contact plate methods), and testing of water-miscible cosmetics.

The presence of other bacteria and food sample components may hinder growth and recovery of Salmonella spp. Pre-enrichment in a nonselective medium such as Nutrient Broth allows for cell damage repair, dilutes toxic or inhibitory substances, and provides a nutritional advantage to Salmonella over other bacteria. Mueller Hinton Agar II is used in antimicrobial susceptibility testing by the disk diffusion method. This formula conforms to Clinical and Laboratory Standard Institute (CLSI), formerly National Committee for Clinical Laboratory Standards (NCCLS).
Mueller Hinton Agar II is based on the formula recommended by Mueller and Hinton for the primary isolation of Neisseria species. Mueller and Hinton selected pea meal extract agar as a simple transparent medium containing heat stable ingredients. During their modification, starch replaced the growth-promoting properties of pea extract, acting as a “protective colloid” against toxic substances.
Bauer, Kirby, Sherris and Tuck recommended Mueller Hinton Agar for performing antibiotic susceptibility tests using a single disk of high concentration. This un-supplemented medium has been selected by the Clinical and Laboratory Standard Institute (CLSI) for several reasons. This medium is low in sulfonamide, trimethoprim and tetracycline inhibitors, and provides satisfactory growth of most non-fastidious pathogens along with demonstrating batch-to-batch reproducibility.
Mueller Hinton Agar is often abbreviated as M-H Agar and complies with requirements of the World Health Organization. Mueller Hinton Agar is specified in FDA Bacteriological Analytical Manual for food testing, and procedures commonly performed on aerobic and facultatively anaerobic bacteria. A variety of supplements can be added to Mueller Hinton Agar, including 5% defibrinated sheep or horse blood, 1% growth supplement and 2% sodium chloride.

LB Broth (Miller) is used in molecular genetic studies in a laboratory setting. LB Broth, Miller is not intended for use in the diagnosis of disease or other conditions in humans. LB Broth (Miller) is based on the formula described by Miller. This medium is used for the growth and maintenance of Escherichia coli strains used in molecular microbiology procedures. LB Broth, Miller is a nutritionally rich medium designed for growth of pure cultures of recombinant strains. E. coli is grown to late log phase in LB Medium. Some plasmid vectors replicate to high copy numbers without selective amplification. Some vectors do not replicate so freely and need to be selectively amplified. Chloramphenicol can be added to inhibit host synthesis and prevent replication of the bacterial chromosome. This broth contains a high level of sodium chloride to aid the maintenance of plasmids. If working with temperate bacteriophages, such as lambda, the addition of magnesium sulphate at 2g/l is recommended to promote phage absorption. If required the medium may be aseptically supplemented with glucose after sterilization.

LB Broth formula is based on L Broth described by Lennox for growth and maintenance of E. coli strains used in molecular microbiology procedures. LB Broth is nutritionally rich, formulated for the isolation of pure recombinant strains. E. coli is grown to late log phase in LB Broth. LB Broth contains ten times the sodium chloride level of Luria Broth, Miller and one half of that found in LB Broth, Miller. This permits the researcher to select the optimal salt concentration for a specific strain. If desired, the medium may be aseptically supplemented with glucose to prepare the complete medium described by Lennox.
HC Agar Base is used with Tween 80 (Polysorbate 80) for the enumeration of molds in cosmetics in a laboratory setting. HC Agar Base is not intended for use in the diagnosis of disease or other conditions in humans.
Methods for isolating molds from cosmetic products require incubation for 5 – 7 days using traditional agar media. In 1986, Mead and O’Neill described a new medium, HC Agar, for enumerating molds in cosmetic products. The formulation of HC Agar decreased incubation time to 3 days at 27.5 ± 0.5°C for molds. HC Agar Base, based on the HC Agar formula of Mead and O’Neill, is supplemented with Tween 80 (Polysorbate 80) to prepare HC Agar.

This buffer is also referred to as Butterfield’s Buffered Phosphate Diluent and recommended for examination of food. Phosphate Buffer, pH 7.2 stabilizes the pH of water used for dilutions.

D/E Neutralizing Agar with Tween is used for the isolation of microorganisms from sanitized environmental surfaces and is not intended for use in the diagnosis of disease or other conditions in humans.
D/E Neutralizing Agar with Tween was developed by Dey and Engley to neutralize a broad spectrum of disinfectants and preservative antimicrobial chemicals, including quaternary ammonium compounds, phenolics, iodine, chlorine preparations, mercurials, formaldehyde, and glutaraldehyde. D/E Neutralizing media neutralize higher concentrations of residual antimicrobials when compared with other standard neutralizing formulations, such as Letheen media, Thioglycolate media, and Neutralizing Buffer.
Total neutralization of disinfectants is critical. Disinfectant residues can result in a false negative (nogrowth) test. D/E Neutralizing Agar effectively neutralizes the inhibitory action of disinfectant carryover, allowing differentiation between bacteriostasis and true bactericidal action of disinfectant chemicals. This is a critical characteristic to consider when evaluating a disinfectant. D/E Neutralizing Agar with Tween is recommended for use in disinfectant evaluations, environmental sampling (swab and contact plate methods), and testing of water-miscible cosmetics.

Dichloran Glycerol (DG-18) Agar Base is used for the selective isolation and enumeration of yeasts and molds from foods in a laboratory setting. Dichloran Glycerol (DG-18) Agar Base is not intended for use in the diagnosis of disease or other conditions in humans. Dichloran Glycerol (DG-18) Agar Base is used for the enumeration of viable osmophilic yeasts and xerophilic molds in food or animal feed products with a water activity of less than or equal to 0.95. This includes such foods as dry fruits, jams, cakes, dried meat, salted fish, grains, cereals, flours, nuts, spices, condiments and some animal feeds. This medium is not suitable for the examination of dehydrated products with a water activity of greater than 0.95, Dichloran Rose Bengal Chloramphenicol Agar (NCM0082 or NCM0029) should be used. The reduction in water activity in this medium is achieved by the addition of glycerol at approximately 18% and this is very important as many yeast and molds require a low water activity to enhance growth and colony development. The medium also contains the antifungal agent dichloran, which restricts the spreading of mucoraceous fungi and restricts the colony size of other genera making colony counting an easier task. Additional selectivity against bacterial growth is achieved by the incorporation of the heat-stable antibiotic Chloramphenicol. Glucose is incorporated as the fermentable carbohydrate source, with casein enzymatic digest providing the essential vitamins, minerals, amino acids, nitrogen and carbon.
