Pseudomonas Isolation Agar is based on Medium A described by King, Ward, and Raney. This medium is very useful for isolating Pseudomonas spp. Pseudomonas Isolation Agar includes Irgasan®, a potent broad spectrum antimicrobial not active against Pseudomonas spp. This medium is selective and formulated to enhanced formation of blue or blue-green pyocyanin pigment by Pseudomonas aeruginosa. The pigment diffuses into the medium surrounding growth.
There are two types of solid culture media for the primary isolation of mycobacteria, coagulated egg as a base (Lowenstein formulations) and an agar base (Middlebrook formulations). The use of agar-based media for primary isolation of mycobacteria have the following significant advantages:

Pseudomonas Agar Base is used for the selective and differential isolation of Pseudomonas sp. using CN or CFC supplementation and is not intended for use in the diagnosis of disease or other conditions in humans.
Pseudomonas Agar Base was developed to be supplemented with CN X107 (Cetrimide & Nalidixic Acid) as per ISO 16266-2:2018, or with CFC X108 (Cetrimide, Fusidate, & Cephaloridine) as per ISO 13720:2010. This base formula is a modification of King’s A Medium.
The CN Supplement is recommended for the isolation of Pseudomonas aeruginosa, where the addition of Nalidixic Acid and the reduction of Cetrimide improved recovery. Pseudomonas Agar Base, with the addition of CN Supplement, demonstrated enhanced pigment formation of Ps. aeruginosa and increased inhibition of Klebsiella, Proteus, and Providencia spp. Pseudomonas Agar Base with the addition of CFC Supplement is recommended for the selective isolation of Pseudomonas spp. Mead and Adams reduced Cetrimide, permitting growth of all pigmented and non-pigmented psychrophilic pseudomonads. The antimicrobics, Fusidate and Cephaloridine, were added to increase the selectivity of the medium. This formulation is used for Pseudomonas spp. from chilled foods and processing plants.
Pseudomonas Agar Base is recommended by ISO for the enumeration of Pseudomonas spp. from meat and meat products according to ISO 13720, and for the detection of enumeration of Pseudomonas aeruginosa spp. in water according to ISO 16266.

mCASE utilizes a dual chromogenic system to differentiate between Salmonella and non-target organisms that grow on the agar. The first chromogen is a target for esterase activity present in all Salmonella species, and results in blue/green colonies. The second chromogen is a target for β-glucosidase activity, present in non-target organisms that are able to grow on the agar and also possess esterase activity. Utilization of the second chromogen masks the presentation of the first, resulting in black colonies in non-Salmonella organisms. All other organisms are either inhibited or grow colorless on the agar. The medium contains an opacity agent that provides a white background, facilitating easy reading and identification. The formulation is the result of modification of CASE (NCM1006), allowing for improved recovery and detection of S. Typhi and S. Paratyphi. The media is able to detect non-motile Salmonella as well as monophasic variants. The media is also able to detect serovars that traditionally present weak esterase activity), and lactose positive strains. Sub-culture should be performed from a selective enrichment.
mCASE is not intended for use with food testing, instead CASE (NCM1006) should be used.

XLT4 Agar is used with Tergitol 4 for the isolation of non-typhi Salmonella spp and is not intended for use in the diagnosis of disease or other conditions in humans. XLT4 Agar is a selective differential isolation medium for the specific detection of Salmonella spp. from environmental and food samples. Due to its highly selective nature, XLT4 Agar is particularly effective when used with samples where overgrowth of contaminating flora is expected, for example, fecallycontaminated agricultural samples. Developed to perform as per Miller & Tate in 1990, this medium was found to improve the recovery of non- typhi Salmonella from chicken and farm environmental samples. Dusch and Altwegg further established the application of XLT4 Agar to salmonellae detection in clinical samples, with the notable exceptions of Salmonella typhi and Salmonella paratyphi. The presence of peptone and yeast extract provides sufficient nutrients to allow the optimal growth of Salmonella spp. Selectivity is provided by the anionic surfactant Niaproof® 4 (formerly known as Tergitol-4 / sodium tetradecylsulfate). This compound acts as an effective selective agent which is active against Grampositive and many Gram-negative organisms, including Proteus spp. Differentiation is based on fermentation of the sugars xylose, lactose and sucrose in addition to the decarboxylation of lysine. The inclusion of the pH indicator, phenol red, provides visual evidence of a pH decrease (yellow) or increase (red) in the medium. Ammonium iron (III) citrate is present to distinguish hydrogen-sulfide (H2S) producing from non-H2S producing organisms. Most enteric organisms, except Shigella, will ferment xylose to produce acid. However, the salmonellae will also decarboxylate the lysine to keep the pH neutral to alkali, thus maintaining red coloration. At nearneutral pH Salmonella can produce H2S from the reduction of ammonium iron (III) citrate and thiosulphate ions producing black or black-centered colonies. Non H2S-producing salmonellae will be red without a black center. Other Enterobacteriaceae (non-salmonellae) which are not inhibited by Niaproof-4, will ferment xylose, lactose and/or sucrose but will not decarboxylate lysine. This fermentation activity causes a decrease in pH, resulting in a color change within the colonies from red to yellow.

Buffered Peptone Water (BPW) is used for the non-selective pre-enrichment of Salmonella spp. from food and is not intended for use in the diagnosis of disease or other conditions in humans. Edel and Kamelmacher found that food preservation techniques involving heat, desiccation, preservatives, high osmotic pressure, or pH changes cause sublethal injury to Salmonella spp. Preenrichment in a non-selective medium allows for repair of cell damage and facilitates the recovery of Salmonella. Lactose Broth is frequently used for this purpose, but it may be detrimental to recovering Salmonellae. Buffered Peptone Water maintains a high pH over the pre-enrichment period and allows in repair of injured cells that may be sensitive to low pH. This is particularly important for vegetable specimens which have a low buffering capacity. Buffered Peptone Water is used in standard methods.

Muller-Kauffmann Tetrathionate-Novobiocin Broth is used for the selective and differential isolation of Salmonella spp. in a laboratory setting and is included in ISO 6579:2017. This medium is not intended for use in the diagnosis of disease or other conditions in humans. A selective enrichment medium for the detection of Salmonella spp. in food, animal feed and in environmental samples from the food production area as described in ISO 6579-1:2017. Mueller originally described a tetrathionate broth that inhibited lactose fermenting Enterobacteriaceae but not Salmonella. Kauffmann later modified the formulation to include ox bile and brilliant green to improve selectivity. Finally, Jeffries added Novobiocin to inhibit Proteus species. Meat extract and casein provide a source of nitrogen and amino acids and sodium chloride maintain the osmotic balance. Ox bile and brilliant green act as selective agents against non-target microorganisms. Tetrathionate is generated from the sodium thiosulfate. Iodine and calcium carbonate buffer the sulfuric acid generated from tetrathionate reduction. According to ISO 6579-1:2017 subculture is performed from Buffered Peptone Water (BPW) into MKTTn, followed by subculture onto Xylose-Lysine Deoxycholate (XLD) Agar ISO and second agar. Magnesium chloride and malachite green act as selective agents against non-target microorganisms. The formulation is very hygroscopic and will produce a slight exothermic reaction when mixed with water. Tetrathionate Broth Base, Hajna, is recommended in the Compendium of Methods for the Microbiological Examination of Foods and the FDA/BAM for the Environmental Sampling and Detection of Salmonella Enteritidis (SE) in Poultry Houses and whole eggs.. Salmonella organisms may be present but injured in food-processing procedures, which include exposure to low temperatures, sub-marginal heat, drying, radiation, preservative, and sanitizers. Numerous reported cases of Salmonella enteritidis (SE) infections are associated with contaminated shell eggs. SE infections can be very serious and symptoms can include diarrhea, fever, abdominal cramps, headache, nausea and vomiting. Eggs can become contaminated on the farm because a laying hen can become infected with SE and pass the bacteria into the egg before it is laid. If the egg is not refrigerated, the bacteria can grow inside the uncracked, whole egg. Tetrathionate Broth Base, Hajna was developed by Hajna and Damon.

Modified Semi-Solid Rappaport-Vassiliadis (MSRV) Agar is used with Novobiocin (NCM4040) for the rapid detection of motile Salmonella spp. and is not intended for use in the diagnosis of disease or other conditions in humans. MSRV was developed in 1986 by De Smedt, Bolderdijk and Rappold as a rapid means of Salmonella detection. The medium, based upon Rappaport Vassiliadis broth, is inoculated directly from the preenrichment medium, in the center of the plate. Motile organisms spread from the center in the semi-solid agar, but non-salmonellas are inhibited by the selective agents. After overnight incubation the use of polyvalent salmonella antisera or a latex kit can confirm the presence of a Salmonella. Alternatively, a paper disc wetted with polyvalent H antiserum can be placed 1/3 of the way from the edge of the dish, and will signal the presence of a Salmonella by inhibiting the mobility of the organism around the disc. Using this medium De Smedt and Bolderdijk have reported the possibility of detecting Salmonella in 24hrs (1987).

Bismuth Sulfite Agar is used for the selective isolation of Salmonella spp. In a laboratory setting. Salmonellosis can result from consumption of raw, undercooked, or improperly processed foods contaminated with Salmonella. U. S. federal guidelines require various poultry products to be routinely monitored before distribution for human consumption. Bismuth Sulfite Agar is a modification of Wilson and Blair formula. The typhoid organism grows luxuriantly on the medium, forming characteristic black colonies. Gram-positive bacteria and coliforms are inhibited on Bismuth Sulfite Agar. The inhibitory action of Bismuth Sulfite Agar permits the use of a large inoculum, increasing the possibility of recovering pathogens that may be present in small numbers. Bismuth Sulfite Agar is generally accepted for routine detection of most Salmonella spp. Bismuth Sulfite Agar is used for the isolation of S. typhi and other Salmonella spp. from food. Kristensen, Lester, and Jurgens first described the use of Brilliant Green Agar as a primary plating medium for the isolation of Salmonella. The report described the medium as useful for the differentiation of “paratyphoid B” from other intestinal gram-negative bacilli. Kaufmann modified their formula and used Brilliant Green Agar in addition to Tetrathionate Broth for the isolation of Salmonella from stool specimens. The outstanding selectivity of this medium permits use of moderately heavy inocula, which should be evenly distributed over the surface. Brilliant Green Agar is used in the microbial limits test. Brilliant Green Agar supplemented with novobiocin is used in food testing.

Brilliant Green Agar is used for the selection and differentiation of Salmonella (other than S. typhi) from foods or feedstuffs. Brilliant Green Agar is not intended for use in the diagnosis of disease or other conditions in humans. First introduced by Kristensen et al in 1925 as a selective medium for the isolation of Salmonella (except S. typhi). The medium was modified by the Netherlands Institute for Public Health, Utrecht. The modification was to increase the selectivity of the medium by increasing the dye concentration. This formulation is quoted by the International Standards Organization, standard European Community Methods, the American Public Health Association and the Association of Official Analytical Chemists. The medium is suitable for subcultures from selective enrichment media. However, because this medium is highly selective, small numbers of salmonellae may be missed. This medium is not recommended for S. typhi and Shigella spp. Less inhibitory media such as X.L.D. (NCM0021) and Hektoen Enteric Agar (NCM0006) will be useful in detecting Salmonella typhi and Shigella spp. which are inhibited by Brilliant Green Agar.

Rappaport Vassiliadis R10 Broth is used for the selective enrichment of Salmonella spp. from foods in a laboratory setting. Rappaport Vassiliadis R10 Broth is not intended for use in the diagnosis of disease or other conditions in humans.
Rappaport et al formulated an enrichment medium for Salmonella spp. that was modified by Vassiliadis et al. The Rappaport formulation, designated R 25/37°C, recommended incubation at 37°C. The Vassiliadis modification, designated R 10/43°C, had a reduced level of Malachite Green and recommended incubation at 43°C. Peterz later showed that incubation at 41.5 ± 0.5°C for 24 hours improved recovery of Salmonella spp. Rappaport-Vassiliadis R10 Broth is a selective enrichment medium that is used following pre-enrichment of the specimen. It has gained approval for use in analyzing milk and milk products, raw flesh foods, highly contaminated foods, and animal feeds. This medium selectively enriches for Salmonella spp. because bacteria, including other intestinal bacteria, are typically susceptible to or inhibited by Malachite Green, high osmotic pressure and/or low pH. S. typhi and S. choleraesuis are sensitive to Malachite Green and may be inhibited.

Xylose Lysine Deoxycholate Agar is used for the isolation and differentiation of enteric pathogens. Conforms to Harmonized USP/EP/JP Requirements. XLD Agar is not intended for use in the diagnosis of disease or other conditions in humans.
A medium recommended by the Harmonized Pharmacopeia for isolation and identification of Salmonella from non-sterile products. Conforms to USP/EP/JP performance specification. Originally formulated by Taylor to differentiate enteric pathogens, the agar is widely used as the preferred differential medium for Salmonella spp. The medium is void of peptones but instead uses yeast extract as a carbon, nitrogen and vitamin source and xylose, lactose and sucrose are fermentable carbohydrates. Salmonella are able to ferment xylose to produce acid but not lactose or sucrose. When the xylose is exhausted Salmonella will decarboxylate lysine shifting the pH back to neutral. At near neutral pH, Salmonella can reduce sodium thiosulfate producing hydrogen sulfide which creates a complex with ferric ammonium citrate to produce black or black centered colonies. Other organisms are able to decarboxylate lysine but acid production from the fermentation of lactose and sucrose keeps the pH too acidic for H2S production. Selectivity is achieved through the incorporation of sodium deoxycholate and phenol red acts as a pH indicator. According to the Harmonized Pharmacopeia, Rappaport Vassiliadis Salmonella Enrichment Broth (NCM0103) is used as a selective enrichment broth, with subculture performed onto Xylose Lysine Deoxycholate (XLD) agar.

Malachite green and magnesium chloride act as selective agents, combined with high osmotic pressure and low pH to effectively inhibit non-target competitive organisms. Some Salmonella (S. typhi and S. enteritidis) are known to be sensitive to malachite green and as such may fail to grow. If these organisms are suspected an alternative selective enrichment broth (e.g. NCM0126 Mueller-Kauffmann Tetrathionate novobiocin Broth – MKTTn) should be used in parallel. This formulation is hygroscopic and will produce a slight exothermic reaction when mixed with water. According to the Harmonized Pharmacopoeia, Rappaport Vassiliadis Salmonella Enrichment Broth is used as a selective enrichment broth, with subculture performed onto Xylose Lysine Deoxycholate (XLD) agar (NCM0027).

Harlequin® Chromogenic Agar for Salmonella Esterase (CASE) is a selective chromogenic agar for the detection of Salmonella that can be used as part of Neogen’s One Broth One Plate Salmonella solution and ISO 6579-1:2017 workflow. It is not intended to diagnose diseases or other conditions in humans.
CASE utilizes a dual chromogenic system to differentiate between Salmonella and non-target organisms that grow on the agar. The first chromogen is a target for esterase activity present in all Salmonella species and results in blue/green colonies. The second chromogen is a target for β-glucosidase activity, present in non-target organisms that are able to grow on the agar and also possess esterase activity. Utilization of the second chromogen masks the presentation of the first, resulting in black colonies in non-Salmonella organisms. All other organisms are either inhibited or grow colorless on the agar. The medium contains an opacity agent that provides a white background, facilitating easy reading and identification. The media can detect non-motile Salmonella (S. Pullorum and S. Gallinarum) as well as monophasic variants (1,4,[5],12:i:-). The media can also detect serovars that traditionally present weak esterase activity (S. Dublin) and lactose-positive strains (S. Arizonae). According to ISO 6579-1:2017, subculture is performed separately from both Rappaport-Vassiliadis medium with Soya (RVS broth) and Muller-Kauffmann tetrathionate-novobiocin (MKTTn)broth.

Tetrathionate Broth Base is not intended for use in the diagnosis of disease or other conditions in humans. Tetrathionate Broth Base is used as a selective enrichment for the cultivation of Salmonella spp. Salmonella organisms may be injured in food-processing procedures, which include exposure to low temperatures, sub-marginal heat, drying, radiation, preservative, and sanitizers. Mueller demonstrated the effectiveness of Tetrathionate Broth for enriching typhoid and paratyphoid bacilli while inhibiting coliform organisms. Using modified Mueller’s broth, Kauffmann increased the number of positive isolates. Tetrathionate Broth was used in studies for the poultry industry and in a collaborative study for rapid screening of Salmonella in food. Tetrathionate Broth Base, abbreviated as TT Broth Base, is specified in standard methods for Salmonella testing. The FDA, Bacteriological Analytical Manual incorporate Tetrathionate Broth Base as a pre-enrichment medium for detecting Salmonella in food materials.

Hektoen Enteric Agar is used for the isolation and differentiation of enteric pathogens. Hektoen Enteric Agar is not intended for use in the diagnosis of disease or other conditions in humans. Hektoen Enteric Agar was developed in 1967 by King and Metzger. Compared to other enteric differentiating media commonly used, Hektoen Enteric Agar increased the isolation rate of Salmonella spp. and Shigella spp. This was accomplished by increasing the carbohydrate and peptone content of the medium in order to counteract the inhibitory effects of bile salts and indicators. King and Metzger formulated a medium that slightly inhibited growth of Salmonella and Shigella, while inhibiting Gram-positive microorganisms. Enzymatic Digest of Animal Tissue provides nitrogen, carbon, and amino acids required for organism growth. Yeast Extract is a vitamin source. Bile Salts Mixture and Acid Fuchsin inhibit Gram-positive organisms. Lactose, Sucrose, and Salicin are fermentable carbohydrates. Sodium Chloride maintains the osmotic balance of the medium. Ferric Ammonium Citrate, a source of iron, allows the detection of hydrogen sulfide (H2S) produced from Sodium Thiosulfate. H2S-positive colonies have black centers. Bromothymol Blue is added as the pH indicator. Agar is the solidifying agent.

Orange Serum Agar is used for the cultivation of aciduric microorganisms associated with spoilage of products and is not intended for use in the diagnosis of disease or other conditions in humans. The low pH of fruit juices makes citrus fruit products susceptible to spoilage by yeasts, molds, and the bacteria Lactobacillus and Leuconostoc. In the 1950’s, Hays investigated spoilage in frozen concentrated orange juice. He found that an agar medium containing orange serum (juice) was superior to Lindegren Agar in isolating the microorganisms responsible for spoilage causing a buttermilk off-odor. Murdock, Folinazzo, and Troy found Orange Serum Agar, pH 5.4 to be a suitable medium for growing Leuconostoc, Lactobacillus, and yeasts. Orange Serum Agar is recommended for examining fruit beverages.

Vogel Johnson Agar is used for the isolation of staphylococci in a laboratory setting. Vogel Johnson Agar is not intended for use in the diagnosis of disease or other conditions in humans.
Coagulase-positive staphylococci, primarily Staphylococcus aureus, are among the microorganisms that cause spoilage or chemical changes in cosmetic products. To isolate coagulase-positive, mannitol fermenting staphylococci, Vogel and Johnson modified Tellurite-Glycine Agar by Zebovitz, Evans, and Niven. The modification included increasing mannitol and adding a pH indicator. Vogel Johnson Agar selects and differentiates coagulase-positive staphylococci that ferment mannitol and reduce tellurite.
Vogel Johnson (VJ) Agar is specified in standard methods testing for cosmetics and nutritional supplements.

An enumeration and detection medium of coagulase-positive Staphylococci from food and animal feeding stuffs using the Most Probable Number (MPN) technique, as described in ISO 6888-3:2003.
Originally described by Giolitti and Cantoni as a medium for the enrichment of staphylococci from foodstuffs, Mossel later applied the medium to use with samples from dried milk and infant food. Optimized for use in samples where staphylococci may be stressed and/or in low numbers, growth of the target organisms is promoted by sodium pyruvate, Glycine and the high concentration of mannitol. Selectivity is achieved via lithium chloride, which inhibits Gram-negative bacilli, and potassium tellurite, which inhibits Gram-positive organisms other than staphylococci. Further selectivity is achieved by use of anaerobiosis either by pouring a plug of agar/paraffin or by incubation in a jar or incubator under anaerobic conditions. Anaerobiosis particularly inhibits the growth of Micrococcus spp. The presence of coagulase-positive staphylococci is indicated by the reduction of tellurite, resulting in a blackening of the broth or a black precipitate. Coagulase-positive staphylococci are principally Staphylococcus aureusbut may also include the species Staphylococcus intermedius and Staphylococcus hyicus. According to ISO 6888-3:2003, subculture is performed separately on Baird Parker Agar or Rabbit Plasma Fibrinogen Agar (RPFA). This medium conforms to the performance and formulation requirements of ISO 6888-3:2003.

Baird-Parker Agar was first described in 1962. It is a selective medium for the isolation and presumptive identification of coagulase-positive staphylococci. This medium is used extensively for detecting Staphylococcus aureus in foods, dairy products, and other materials. Coagulase-positive staphylococci can grow and reproduce in cosmetic products. These products are tested for the presence of coagulase-positive staphylococci using standard microbiological methods.

Mannitol Salt Agar is used for the isolation of staphylococci in a laboratory setting. Mannitol Salt Agar is not intended for use in the diagnosis of disease or other conditions in humans. Conforms to Harmonized USP/EP/JP Requirements. Chapman formulated Mannitol Salt Agar to isolate staphylococci by inhibiting growth of most other bacteria with a high salt concentration. Chapman added 7.5% Sodium Chloride to Phenol Red Mannitol Agar and noted pathogenic strains of staphylococci (coagulase-positive staphylococci) grew luxuriantly and produced yellow colonies with yellow zones. Non-pathogenic staphylococci produced small red colonies with no color change to the surrounding medium. Mannitol Salt Agar is highly selective and samples from heavily contaminated sources may be streaked onto this medium without danger of overgrowth. Mannitol Salt Agar is recommended for isolating pathogenic staphylococci from samples, cosmetics, and microbial limit tests. Bacteria that grow in the presence of a high salt concentration and ferment mannitol produce acid products, turning the phenol red pH indicator from red to yellow. Typical pathogenic staphylococci ferment mannitol and form yellow colonies with yellow zones. Typical non-pathogenic staphylococci do not ferment mannitol and form red colonies. Enzymatic digest of casein, enzymatic digest of animal tissue, and beef extract provide the nitrogen, vitamins, and carbon in Mannitol Salt Agar. D-Mannitol is the carbohydrate source. In high concentrations, sodium chloride inhibits most bacteria other than staphylococci. Phenol red is the pH indicator. Agar is the solidifying agent.

Baird Parker Agar is used for the cultivation of Staphylococcus aureus from foodstuffs according to ISO 6888, and is not intended for use in the diagnosis of disease or other conditions in humans.
Originally introduced in 1962, this medium was developed by Baird-Parker to overcome the problems of recovering damaged Staphylococcus aureus from foodstuffs. This version of the medium is formulated according to ISO 6888-1:1999 A1:2003 and is in compliance with ISO 6888-2:2003 A1:2003 and ISO 6888-3:2003.
Baird-Parker medium is highly selective by nature, due to the presence of potassium tellurite and lithium chloride. Tellurite inhibits most coliforms and is also reduced to telluride by S. aureus, giving the typical black colonies. Glycine and sodium pyruvate are both used as growth factors by staphylococci while the pyruvate also neutralizes any toxic peroxides that may be formed.
When Baird-Parker medium is used with Egg Yolk Tellurite (NCM4010), presumptive S. aureus appear as black colonies demonstrating lecithinase activity (an opaque zone around the colony) and lipase activity (a zone of clearing encircling the opaque zone). Suspected S. aureus colonies should be confirmed with RPF for coagulase or latex agglutination test.
Rabbit plasma fibrinogen (NCM4052) is a more specific alternative to egg yolk tellurite and allows the direct detection of coagulase-positive S. aureus. Typical S. aureus appear as black colonies surrounded by a zone of precipitation (demonstrating coagulase activity). This is recognized as the gold standard method for the identification of S. aureus. RPF overcomes any issues with atypical colony forms and its use means further confirmatory tests are not necessary.

Todd Hewitt Broth is used for the cultivation of streptococci and other fastidious microorganisms. Todd Hewitt Broth is not intended for use in the diagnosis of disease or other conditions in humans. Todd Hewitt Broth was originally developed for the production of antigenic streptococcal hemolysin. Todd Hewitt Broth is prepared according to the formula described by Updyke and Nickle, who compared media for type specific extract production of group A streptococci. This study was performed using Todd Hewitt Broth prepared with infusion of fresh beef heart as a control. Results showed Todd Hewitt Broth was particularly satisfactory for growth of group A streptococci for serological typing. Todd Hewitt Broth is recommended as an enrichment medium for growth of streptococcal cells in the identification of groups A and B by IF staining. Todd Hewitt Broth was used as an enrichment broth for group A streptococci in a comparison study of a rapid antigen test.

Bile Esculin Azide Agar is used for the selective isolation and differentiation of group D streptococci in a laboratory setting. Bile Esculin Azide Agar is not intended for use in the diagnosis of disease or other conditions in humans.
Bile Esculin Azide Agar is a modification of the medium reported by Isenberg and Isenberg, Goldberg, and Sampson. This formula modifies Bile Esculin Agar by adding sodium azide and reducing the concentration of bile. The revised medium is more selective, but still provides rapid growth and efficient recovery of group D streptococci.
Molecular taxonomic studies of the genus Streptococcus have placed enterococci, previously described group D streptococci, in the genus Enterococcus. The ability to hydrolyze esculin in the presence of bile is a characteristic of enterococci and group D streptococci. Swan compared the use of an esculin medium containing 40% bile salts with the Lancefield serological method of grouping and reported that a positive reaction on the bile esculin medium correlated with a serological group D precipitin reaction. Facklam and Moody found that the bile esculin test provided a reliable means of identifying group D streptococci and differentiating them from non-group D streptococci.
Bile Esculin Azide Agar selected for S. bovis, displayed earlier distinctive reactions, and eliminated the requirement for special incubation temperatures.

Bile Esculin Agar is based on the formulation described by Swan and further evaluated by Facklam and Moody. Rochaix first noted the value of esculin hydrolysis in the identification of enterococcci. Meyer and Schonfeld added bile to the esculin medium and demonstrated 61 of 62 enterococci strains were able to grow and hydrolyze esculin, while other streptococci could not.
