Fastidious Anaerobe Agar is used for the cultivation of anaerobic microorganisms and is not intended for use in the diagnosis of disease or other conditions in humans. A primary isolation medium capable of growing most clinically significant anaerobes. Developed by Lab M (Neogen® Corporation), comparisons have shown this medium to be superior to other formulations as a primary isolation medium for fastidious organisms. The peptones included have been chosen for maximum growth stimulation. Starch and sodium bicarbonate act as de-toxification agents while hemin encourages pigment production in Porphyromonas melaninogenicus. Specific growth promoting agents are Cysteine for Fusobacterium necrophorum, Propionibacterium acne and Bacteriodes fragilis, arginine for Eubacterium spp. soluble pyrophosphate for Porph. gingivalis and Porph. asaccharolyticus. Pyruvate helps neutralize hydrogen peroxide and is also utilized by Veillionella spp. as an energy source. Vitamin K and sodium succinate provide essential growth factors for some anaerobes as does the 0.1% glucose. The low level of glucose prevents the production of high levels of acids and alcohols which would inhibit colonial development.

Fastidious Anaerobe Broth (F.A.B.) was developed by Neogen, working in conjunction with the microbiology department of a University of Manchester teaching hospital. The medium was designed to give optimum growth of fastidious anaerobes and has found applications as a blood culture medium and an enrichment broth for the isolation of anaerobes. The medium is very rich in nutrients from the specially selected peptone mixture. Vitamin K. hemin and L-cysteine are all growth factors required by some anaerobes. L-cysteine together with sodium thioglycolate reduce the Eh of the medium and the agar content inhibits absorption of oxygen and convection currents. Resazurin is a redox indicator. Several published evaluations show F.A.B. to be the liquid medium of choice for fastidious anaerobes.

Quastel and Stephenson found the presence of small amounts of a compound containing an –SH group (cysteine, thioglycollic acid, and glutathione) permitted “aerobic” growth of Clostridium sporogenes. Falk, Bucca, and Simmons discovered the advantages of using small quantities of agar in detecting contaminants during sterility testing. Brewer demonstrated the value of a small amount of agar and a reducing substance in this medium.
Thioglycolate Medium w/o Indicator is used for cultivating and detecting microorganisms in normally sterile materials, especially those containing mercurial preservatives when the oxidation-reduction indicator is not present or required. Thioglycolate Medium w/o Indicator is the medium of choice for diagnostic testing, where lack of an indicator avoids possible toxicity to organisms.

Iron Sulphite Agar is a medium for the detection of thermophilic anaerobic organisms causing sulphide spoilage in food, and is not intended for use in the diagnosis of disease or other conditions in humans.
This formulation is a modification of Cameron Sulphite Agar, which was developed by the National Canners Association of America (now the Grocery Manufacturers Association). Iron Sulphite Agar has a reduced concentration of sodium sulphite to allow improved detection of some strains of Clostridium sporogenes. Beerens, and later Mossel, demonstrated that some strains of C. sporogenes would not tolerate sodium sulphite levels of 0.1%. Mossel further observed that reducing sulphite content to 0.05% improved detection of these strains.
Tryptone provides nitrogen and other nutrients necessary to support bacterial growth. The presence of sulphite reducing bacteria is indicated by the formation of black colonies. These colonies form when bacteria reduce sulphite to sulphide, which reacts with iron (III) citrate to yield a black precipitate.

Tryptone Soy Broth (TSB) is a general-purpose broth which will support the growth of a wide range of micro-organisms and is not intended for use in the diagnosis of disease or other conditions in humans.
Tryptic Soy Broth, a general purpose medium, is commonly referred to as Casein Soya Bean or Soybean- Casein Digest Broth, and abbreviated as TSB. This medium was originally developed for use without blood in determining the effectiveness of sulfonamides against pneumococci and other organisms. Clostridia and non-sporulating anaerobes grow luxuriantly in this broth when incubated under anaerobic conditions. TSB is recommended for testing bacterial contaminants in cosmetics and complies with established standards in the food industry.
Enzymatic Digest of Casein and Enzymatic Digest of Soybean Meal are nitrogen sources in TSB. Dextrose is the carbon energy source that facilitates organism growth. Sodium Chloride maintains osmotic balance; Dipotassium Phosphate is a buffering agent.

Holbrook and Anderson described a highly selective and diagnostic medium (PEMBA) for the isolation and determination of Bacillus cereus from food. The medium is formulated to detect small numbers of B. cereus in the presence of large number of contaminants. This medium differentiates B. cereus from other bacteria based on resistance to polymyxin, lack of mannitol fermentation, and presence of lecithinase. B. cereus can be found on vegetables, processed foods, and in nature. B. cereus causes gastrointestinal illness if the organism is allowed to proliferate. Outbreaks of foodborne illness have been associated with boiled and cooked rice, cooked meats, and cooked vegetables.

Bacillus Cereus Mannitol Egg Yolk (MYP) Agar is used with supplements for the selective and differential enumeration of Bacillus cereus from foods. NCM0062 conforms to ISO and FDA / BAM formulation. MYP Agar Base is not intended for use in the diagnosis of disease or other conditions in humans.
Formulated according to ISO 7932:2004, MYP was introduced by Mossel and his co-workers in 1967 for the enumeration of Bacillus cereus in foods and was shown to be the most effective for this purpose by Inal in 1972. B. cereus is differentiated from other members of the Bacillus group by two reactions: mannitol fermentation and lecithinase production. Mannitol fermentation on this medium produces a yellow color, due to the indicator dye, phenol red changing color as acid is produced. B. cereus is mannitol negative and produces red colonies. The lecithinase production of B. cereus is indicated by a white precipitate around the colonies. Polymyxin is added to suppress coliforms but some Proteus spp. and Gram-positive cocci may grow.

Universal Beer Agar is used for the cultivation of bacteria and yeasts encountered in the brewing industry in a laboratory setting. Universal Beer Agar is not intended for use in the diagnosis of disease or other conditions in humans.
Universal Beer Agar was developed by Kozulis and Page and used as a basal medium supplemented with beer. The developers compared this formula with other media commonly used in breweries for detecting microbial contamination. The characteristics of Universal Beer Agar are closer to natural environmental conditions found in a typical brewery. Universal Beer Agar supports the growth of more lactic acid bacteria and yields larger colonies in a shorter time than traditional brewer’s media. Supplementing this medium with beer has produced a selective environment for organisms that are adapted to existent conditions in the brewery. The presence of hop constituents and alcohol inhibits growth of many airborne microorganisms not adapted to this environment.
Universal Beer Agar supports growth of Lactobacillus, Pediococcus, Acetobacter, and yeasts that are known beer contaminants. Universal Beer Agar is abbreviated as UBA.

Campylobacter Blood-Free Selective Medium (Modified Charcoal Cefoperazone Deoycholate Agar or mCCDA) is used for the cultivation of campylobacters, and is not intended for use in the diagnosis of disease or other conditions in humans.
A blood free medium which will support the growth of most enteric campylobacters. The selective supplement X112 (X212) makes the medium selective for C. jejuni. and C. coli when incubated at 37°C. With this product incubation at 42°C is no longer necessary and higher recovery rates have been reported at 37°C than at 42°C. The supplement X112 (X212) consists of cefoperazone and amphotericin and is superior to the selective combination of Skirrow, Butzler and Blazer-Wang all of which contain antibiotics shown to be inhibitors to C. coli. The colonial morphologies of Campylobacter spp. on this medium are distinctive.

Eosin Methylene Blue Agar Levine is used for the isolation and differentiation of Gram-negative enteric bacilli and is not intended for use in the diagnosis of disease or other conditions in humans.
This medium was introduced in 1916 by Holt-Harris and Teague to differentiate Escherichia spp. and Aerobacter spp. It was modified by Levine in 1918 who removed sucrose from the formula and increased the lactose content. The distinctive metallic sheen produced by E. coli on this medium is due to acid production resulting in an amide bonding between the eosin and methylene blue, other coliforms do not produce enough acid to cause this reaction. Eosin inhibits most Gram-positive organisms. The prepared medium is sensitive to light.

Differential Reinforced Clostridial Medium ISO (DRCM) is a medium for the detection and enumeration of the spores of sulfite-reducing anaerobes as described in ISO 26461-1, and is not intended for use in the diagnosis of disease or other conditions in humans. Sulfite reducing anaerobes, in particular Clostridia, can be indicators of remote and intermittent pollution. Widespread in the environment, being found in human and animal feces, soil and waste water, the spores are more resistant to physical and chemical factors than vegetative cells and able to survive for long periods in water. The spores may also be resistant to chlorination at the levels commonly used in water treatment. DRCM has been developed for use with the Most Probable Number (MPN) method to determine the MPN of anaerobes (Clostridia) per volume of sample. The formulation includes peptone, yeast extract, meat extract, starch & L-Cysteine for nutrition with glucose providing the energy source. Sodium acetate provides partial selectivity. Clostridia are able to reduce sulfite to sulfide – forming iron sulfide. Iron (III) citrate is included in the formulation as an indicator of sulfite reduction. Blackening in the medium indicates that iron sulfide has been formed and therefore that sulfite reduction has occurred. Other bacteria are able to form sulfide, so vegetative cells must be first be removed from the test sample by an appropriate process e.g. heat treatment.

Fluid Thioglycolate Medium is recommended by the Harmonized Pharmacopoeia for sterility testing and conforms to USP/EP/JP performance specification. In this formulation casein and yeast extract provide a source of nitrogen, essential vitamins and amino acids. The glucose provides a carbon source and sodium chloride maintains osmotic balance. L-Cystine and sodium Thioglycolate act as reducing agents to create an anaerobic environment and maintain a low Eh. This is aided by the low level of agar which reduces the oxygen permeability through the medium. Resazurin is an oxidation indicator which turns from colorless to red/pink when oxidised. Sodium Thioglycolate also serves to inactivate mercurial compounds. If after sterilization more than the upper one third of the medium has become red/pink it may be restored once by heating in a water bath or in free-flowing steam until the color disappears. Ensure the media is cooled quickly and prevent the introduction of non-sterile air into the containers.

Clostridium Difficile Agar Base (Brazier’s) is used with blood, Cycloserine, and Cefoxitin for the isolation of Clostridium difficile. Clostridium Difficile is not intended for use in the diagnosis of disease or other conditions in humans.
Brazier’s CCEY agar is the formulation currently used by the Anaerobe Reference Unit for the isolation of C. difficile, resulting from work initiated by Ken Phillips and Paul Levett, and completed by Jon Brazier. Based upon the market leading anaerobe medium, Fastidious Anaerobe Agar, it incorporates additional ingredients to improve the isolation and differentiation of C. difficile from samples.
Cholic acid is present to promote spore germination following alcohol shock treatment, and p-hydroxyphenylacetic acid to enhance the production of p-cresol, a distinctive metabolite of C. difficile. Selectivity is achieved by addition of supplement X093 (cefoxitin cycloserine) and egg yolk emulsion X075 is added to help differentiate C. difficile from lecithinase positive clostridia. Membrane Lactose Glucuronide Agar (mLGA) is a medium for the detection and identification of E. coli and coliforms from water samples and is not intended for use in the diagnosis of disease or other conditions in humans. Traditionally, membrane Lauryl Sulphate Broth (mLSB) has been used as the standard media for isolating coliforms (including E. coli) from water potentially contaminated with sewage. Harlequin membrane Lactose Glucuronide Agar (mLGA) is a modification of mLSB aimed at reducing costs by reducing the number of filters used per test sample and aiding in the recovery and identification of coliforms and E. coli. The medium has been modified from the mLSB formulation by the incorporation of X-glucuronide (BCIG), sodium pyruvate and agar. X-glucuronide is a chromogenic substrate which detects the β-glucuronidase enzyme – highly specific for E. coli* – and allows for the presumptive isolation of E. coli. Sodium pyruvate aids the recovery of chlorine stressed organisms and agar is incorporated to remove the need for absorbent pads. This medium is recommended for the enumeration of coliform bacteria and E. coli by a single membrane filtration technique in The Microbiology of Drinking Water 2009 (previously Report 71). *96-97% of E. coli strains positive. A notable exception is E. coli O157:H7.

A-1 Medium is used for the detection of coliform organisms in water and food in a laboratory setting. A-1 Medium is not intended for use in the diagnosis of disease or other conditions in humans.
Since the early 1900’s enumeration of coliform organisms, specifically E. coli, have been used to determine water purity. Elevated temperature and most-probable-number (MPN) methods are routinely used for analysis of water and food samples for presence of fecal coliforms. A-1 Medium was formulated to hasten E. coli recovery and reduce the incidence of false positive cultures.
In 1972 Andrews and Presnell developed A-1 Medium. This medium recovers E. coli from estuarine water in 24 hours instead of 72 hours, and in greater numbers without the pre-enrichment step. A-1 Medium can be used in a single step procedure for the detection of fecal coliforms in source water, seawater, treated wastewater, and foods. A-1 Medium conforms to standard methods for the isolation of fecal coliforms in water and foods.

Presence Absence Broth is used for the detection of coliform bacteria in water treatment plants or distribution systems using the presence-absence coliform test in a laboratory setting. Presence Absence Broth is not intended for use in the diagnosis of disease or other conditions in humans.
The Presence Absence (PA) test is a presumptive detection for coliforms in water. The test is a simple modification of the multiple-tube procedure. One 100 mL test sample is inoculated into a single culture bottle to obtain qualitative information on the presence or absence of coliforms, through the presence or absence of lactose fermentation. This test is based on the principle that coliforms and other pollution indicator organisms should not be present in a 100 mL water sample.
Comparative studies with the membrane filter procedure indicate the PA test may maximize coliform detection in samples containing many organisms that could overgrow coliform colonies and cause problems in detection. The PA test is described in standard methods for water testing and U.S. EPA.

Membrane Lauryl Sulphate Broth is used for the enumeration of coliforms in water and is not intended for use in the diagnosis of disease or other conditions in humans.
This medium superseded Membrane Enriched Teepol Broth when Shell Chemicals withdrew Teepol 610 from sale. Sodium lauryl sulphate was found to be an adequate reproducible substitute and this medium is recommended for the enumeration of coliform and organisms in water and sewage.

Lactose Broth is used for the cultivation of Salmonella and coliform bacteria from food, dairy, and water products in a laboratory setting. Lactose Broth is not intended for use in the diagnosis of disease or other conditions in humans. Lactose Broth is frequently used as a pre-enrichment medium when testing foods and dairy products for Salmonella spp. In dried or processed foods, Salmonella species may be sub-lethally injured and in low numbers. The presence of other bacteria as well as components of the food sample may hinder growth and recovery of Salmonella. Pre-enrichment in a non-selective medium such as Lactose Broth allows for repair of cell damage, dilutes toxic or inhibitory substances, and provides a nutritional advantage to Salmonella over other bacteria. Lactose Broth is widely used and is included in many procedures for testing foods, dairy products and other materials. Lactose Broth is also used for the detection of coliform organisms in water, dairy products, and other materials.

Violet Red Bile Agar is used for the enumeration of coliforms in food and dairy products in a laboratory setting. Violet Red Bile Agar is not intended for use in the diagnosis of disease or other conditions in humans and conforms to American Public Health Association (APHA).
The coliform group of bacteria includes aerobic and facultative anaerobic, gram-negative, non-spore forming bacilli. Coliforms ferment lactose and form acid and gas at 35°C within 48 hours. Members of Enterobacteriaceae comprise the majority of the group, but other lactose fermenting organisms may also be included.
Procedures to detect, enumerate, and presumptively identify coliforms are used in testing foods and dairy products. One method for performing the presumptive test for coliforms uses Violet Red Bile Agar, (VRBA). If typical coliform colonies appear, they are tested further to confirm their identification as coliforms. Coliform colonies lower the pH of the medium, subsequently causing their colonies to look red (Neutral Red Dye) and to precipitate the bile salts.
