E. coli / Coliform Agar is for the simultaneous enumeration of Escherichia coli and coliforms in food and environmental samples and is not intended for use in the diagnosis of disease or other conditions in humans.
This dual chromogenic substrate medium makes the different colony types easy to distinguish allowing rapid counting of both E. coli and coliforms on a single agar.
Based upon the formulation of Tryptone Bile Agar, the medium has been modified by the addition of two chromogenic substrates, one to detect the ß-glucuronidase enzyme (X- glucuronide) and another to detect the ß-galactosidase enzyme (magenta-ß-gal). Typical E. coli strains possess both enzymes but only cleave the X-glucuronide substrate, thereby producing blue-green colonies. Typical coliforms, however, possess only the ß-galactosidase enzyme and produce rose-pink colonies.
The colony types are easily distinguishable, even in the presence of other organisms, or when large numbers are observed, making simultaneous enumeration of E. coli and coliforms a quick and simple procedure.

Violet Red Bile Agar W/ MUG is specified in many procedures to enumerate coliforms in food and dairy products. In 1982, Feng and Hartman developed a rapid fluorogenic assay for Escherichia coli by incorporating 4-methylumbelliferyl-β-D-glucuronide (MUG) into Lauryl Tryptose Broth. Similarly, incorporating MUG into Violet Red Bile Agar permits the detection of E. coli among the coliform colonies. Standard methods procedures specify Violet Red Bile Agar with MUG for detecting E. coli in food and dairy products by fluorescence. This medium is often abbreviated as VRBA w/ MUG.

DC Medium with BCIG is used in the chromogenic differentiation of E. coli from other coliforms in water samples using the membrane filtration method in a laboratory setting. DC Medium with BCIG is not intended for use in the diagnosis of disease or other conditions in humans.
DC (Differential Coliform) Medium with BCIG is a selective and differential medium for the presumptive identification of E. coli. This medium is enhanced by the addition of a chromogenic agent, BCIG, 5-bromo-4-chloro-3-indolyl-ß-D-glucuronide to detect glucuronidase activity. The presence of the enzyme ß-D-glucuronidase differentiates most E. coli spp. from other coliforms, and is the same enzyme used in the MUG reaction. BCIG reacts slightly differently, and when released into the medium is insoluble, accumulating within the cell, imparting a blue to purple color to presumptive E. coli colonies.

Peptone and yeast extract provide a source of nitrogen and essential vitamins. Sodium chloride maintains the osmotic balance and the phosphates buffer the media. Sodium Pyruvate, Sorbitol and Tryptophan aid in the recovery and detection of the target organisms. Tergitol™ 7 is a surfactant that acts as a selective agent. The dual chromogens are incorporated to differentiate between E. coli and coliforms based on their enzymatic activity. IPTG induces β-D-galactosidase and agar is the gelling agent. E. coli possess both β-D-galactosidase and β-glucuronidase activity so appear dark blue to violet. Coliform bacteria that are not E. coli possess only β-D-galactosidase so appear pink to red. Any organism able to grow that does not belong to these groups appears colorless. This medium conforms to the performance requirements of ISO 9308-1 2017.

Cystine Lactose Electrolyte Deficient (CLED) Agar is used for the differentiation and enumeration of microorganisms in urine. A medium for urine culture where the absence of electrolytes inhibits the swarming of Proteus spp. Cystine is added for the benefit of those organisms which have a specific cystine requirement. Lauryl Tryptose (LST) Broth is used for the detection of coliform bacteria in water and wastewater. Lauryl Tryptose Broth is not intended for use in the diagnosis of disease or other conditions in humans.
Lauryl Tryptose Broth is a selective medium for the detection of coliforms in water, dairy products and other foods. The American Public Health Authority (APHA) recommend Lauryl Tryptose Broth for the Most Probable Number Presumptive Test of coliforms in waters, effluent or sewage and as a confirmation test of lactose fermentation with gas production from milk samples and for the detection of coliforms in foods.
Lauryl Tryptose Broth is prepared according to the formulation of Mallmann and Darby. Mallmann and Darby showed that tryptose at a concentration of 2% increased the early logarithmic growth phase when compared to meat peptone. These researchers added phosphate buffers and sodium chloride, which improved gas production by “slow lactose fermenting” organisms. Sodium lauryl sulfate was incorporated as a selective agent for the inhibition of non-coliform organisms.
This medium can also be used with the addition of MUG (4- methylumbelliferyl-β-D-glucuronide) according to the ISO Standard 11866-2 to give enhanced detection of Escherichia coli.

EC Broth (Escherichia coli Broth) is a selective enrichment broth designed for the isolation of coliforms, including E. coli, from water and food samples. It was the recommended medium of the American Public Health Association (APHA) and the AOAC.

Organisms that reduce the sodium thiosulfate produce a black precipitate due to the presence of thiosulfate and ammonium iron(III) citrate. Cronobacter spp. possess α-D-glucosidase so appear blue to green. Similar Enterobacteriaceae do not express this enzyme thus grow colorless. According to ISO 22964:2017, subculture is performed after selective enrichment in Cronobacter Selective Broth (CSB). Dermatophyte Test Medium is used for the selective isolation of dermatophytic fungi and is not intended for use in the diagnosis of disease or other conditions in humans.
In 1969, Taplin et al. developed this medium for the isolation and recognition of dermatophytic fungi, the causative agent of ringworm from hair, nails, and skin. Dermatophyte Test Medium is preferred for isolation and early recognition of Microsporum, Trichophyton, and Epidermophyton genera because of a distinct color change in the medium. Rapidly-growing species may produce a complete color change in the medium in 3 days. The slower-growing species will change the indicator in longer time periods. Other organisms may grow but can be recognized as non-dermatophytes by lack of a color change. A few organisms, including saprophytes, yeasts, and bacteria are capable of changing the medium from red to yellow, but are easily recognized by their distinctive colonial morphology.

Harlequin® Tryptone Bile Glucuronide Agar (TBX) is used for the enumeration of E. coli in food and animal feed and is not intended for use in the diagnosis of disease or other conditions in humans. Tryptone Bile Glucuronide Agar is recommended by ISO 16649-1:2001, ISO 16649-2:2001 and ISO 16649-3:2015 for enumeration of β-glucuronidase positive Escherichia coli in products intended for human consumption or for the feeding of animals. The medium contains tryptone as a source of nitrogen and amino acids, bile salts act as a selective against Gram-positive organisms and agar is the gelling agent. 5-bromo-4-chloro-3-indolyl-β-D-glucuronide (X-glucuronide) is a chromogen for the detection of the β-D-glucuronidase enzyme which is present in most E. coli* but not other coliforms. The intracellular enzyme present in E. coli is able to break the bond between the substrate and the chromophore which dimerizes is the presence of oxygen to form an insoluble colored compound within the cell. The accumulation of the colored chromophore results in blue colonies on the agar.
*96-97% of E. coli are β-D-glucuronidase positive. A notable exception is E. coli O157:H7

Enterohemorrhagic E. coli (EHEC) are recognized as the primary cause of hemorrhagic colitis (HC) or bloody diarrhea. This infection can also lead to hemolytic uremic syndrome (HUS). Enterohemorrhagic infections are usually food or water borne and have been implicated in undercooked ground beef, raw milk, lunchmeat, water, unpasteurized apple cider, sprouts and vegetables.
Modified Buffered Peptone Water with Pyruvate is recommended in the sample preparation for the screening method following FDA/BAM Enterohemorrhagic E. coli (EHEC) Protocol. After the sample is incubated for five hours in Modified Buffered Peptone Water with Pyruvate, Cefsulodin, Acriflavin and Vancomycin are added, followed by an additional incubation period. EC Medium w/ MUG is used for the fluorogenic detection of Escherichia coli in a laboratory setting. EC Medium w/ MUG is not intended for use in the diagnosis of disease or other conditions in humans.
EC Medium was developed by Hajna and Perry in an effort to improve the methods for the detection of the coliform group and E. coli. This medium consists of a buffered lactose broth with the addition of 0.15% Bile Salts Mixture. Growth of spore-forming bacteria is inhibited by the bile salts, while growth of E. coli is enhanced by its presence. EC Medium w/ MUG is the same formula as EC Medium, with the addition of 4-methylumbelliferyl-ß-D-glucuronide. Feng and Hartman developed a rapid assay for E. coli by incorporating 4-methylumbelliferyl-ß-D-glucuronide (MUG) into Lauryl Tryptose Broth at a final concentration of 100 ɥg/mL. Moburg determined the amount of MUG could be reduced to a final concentration of 50 ɥg/mL without adversely affecting results.
EC Medium w/ MUG is prepared according to the formula specified by US EPA and methods for water and food testing.

MacConkey Agar is used for the isolation and differentiation of Gram-negative enteric bacilli in a laboratory setting. Conforms to Harmonized USP/EP/JP Requirements and FDA/BAM. Sorbitol MacConkey Agar (SMAC-BCIG) is for the isolation of Escherichia coli O157:H7 and is not intended for use in the diagnosis of disease or other conditions in humans. Pathogenicity of Escherichia coli O157:H7, the primary serovar associated with haemorrhagic colitis (HC) and haemolytic uraemic syndrome (HUS), is linked to the production of verocytotoxins (VT1 and VT2). However, it should be noted that not all strains of O157 produce verocytotoxins, and that strains from other serovars can be toxin producers (e.g. O26, O103, O111, O113, O145). E. coli O157 has been associated epidemiologically with food poisoning outbreaks involving beef burgers and cold cooked meats. This medium is a modification of Sorbitol MacConkey Agar (SMAC). The addition of the chromogenic substrate BCIG (5-bromo-4-chloro-3-indoxyl-ß-D-glucuronide) improves the specificity of the medium. E. coli O157:H7 is typically sorbitol negative and ß-glucuronidase negative producing pale translucent colonies on this medium. The majority of other E. coli strains are ß-glucuronidase positive and sorbitol positive (pink/red colonies). A small percentage of E. coli are ß-glucuronidase positive and sorbitol negative and thus appear as blue/green colonies on this medium. Consequently, this medium can distinguish between non-O157 sorbitol negative E. coli and the genuine toxigenic E. coli O157:H7. This reduces the number of unnecessary confirmation tests that are performed. The medium can be made more selective by the addition of Cefixime Tellurite supplement (X161) to prepare CT-SMAC. Most workers recommend the use of CT-supplemented medium alongside unsupplemented medium to ensure maximum isolation of E. coli O157. This medium can also be useful for the detection of other VTEC producing E. coli in conjunction with specifically targeted IMS beads (Captivate™).

MacConkey Agar without Crystal Violet and Salt is used for the isolation and differentiation of Gram-negative enteric bacilli from specimens containing swarming strains of Proteus spp, and is not intended for use in the diagnosis of disease or other conditions in humans. MacConkey Agar is based on the bile salt-neutral red-lactose agar of MacConkey. The original MacConkey medium was used to differentiate strains of Salmonella typhosa from members of the coliform group. Formula modifications improved growth of Shigella and Salmonella strains. These modifications include the addition of 0.5% sodium chloride, decreased agar content, altered bile salts, and neutral red concentrations. The formula modifications improved differential reactions between enteric pathogens and coliforms. MacConkey Agar without Crystal Violet and Salt is a differential medium that restricts swarming of Proteus spp., aiding in the detection and isolation of enteric microorganisms. Sodium Chloride is deleted from the medium to provide an electrolyte deficient medium preventing Proteus spp. from spreading. Lysine Iron Agar is used for the differentiation of microorganisms on the basis of lysine decarboxylase and hydrogen sulfide production. Lysine Iron Agar is not intended for use in the diagnosis of disease or other conditions in humans. Lysine Iron Agar is prepared according to the formulation of Edwards and Fife, who developed the medium to detect Salmonella arizonae. S. arizonae ferments lactose rapidly, and the authors found expected H2S production on Triple Sugar Iron Agar was suppressed. Detection of S. arizonae is important because it has been implicated in food borne infections. One Plate for Enterobacteriaceae (OP-EBAC) offers a rapid method for the enumeration of total Enterobacteriaceae using traditional culture methodology.
One Plate for Enterobacteriaceae consists of a selective base which permits the detection and enumeration of Enterobacteriaceae, by their ability to ferment glucose and produce acid visualized by an indicator dye. Non-target organisms are inhibited by a mixture of selective compounds incorporated into the base.
The product can permit a quantitative result for Enterobacteriaceae in 18-22 hours for all food products, using only one plate versus 2 or more as described in ISO 7218 and ISO 21528-2:2017. Neither overlays nor confirmations are required for the method.

Glucose OF Medium is used for the confirmation of Enterobacteriaceae in foods according to ISO 21528-1&2:2017, and is not intended for use in the diagnosis of disease or other conditions in humans.
Enzymatic digest of casein provides nitrogen, vitamins, minerals and amino acids essential for growth. Sodium chloride supplies essential electrolytes for transport and osmotic balance. Dipotassium phosphate acts as a buffer system. Glucose is the fermentable carbohydrate providing carbon and energy. Bromothymol blue is the pH indicator.
Colonies of presumptive Enterobacteriaceae should be confirmed by means of tests for the fermentation of glucose and the presence of oxidase. If the colonies are oxidase-negative and glucose-positive, the sample shall be regarded as being positive for Enterobacteriaceae. ISO 21528 :2017 recommends this medium to confirm the presumptive colonies of Enterobacteriaceae by fermentation test.

MacConkey Agar No. 3 is used for the cultivation of Enterobacteriaceae and is not intended for use in the diagnosis of disease or other conditions in humans. A modification recommended by the W.H.O. and the American Public Health Association for the isolation of Enterobacteriaceae from waters and sewage. The medium has been made more selective than MacConkey’s original formula by the use of crystal violet as well as bile salts. Gram positive organisms will not grow on this medium.

Brain Heart Infusion Agar is used for the cultivation of a wide variety of fastidious organisms and is not intended for use in the diagnosis of disease or other conditions in humans.
A general purpose nutritious agar base. This medium was first used for the isolation of dental pathogens. With the addition of 7% defibrinated blood the medium will support the growth of a wide range of fastidious organisms, the phosphate buffer will help neutralize the acids produced from the utilization of glucose and thus maintain viability. The medium is not recommended for the determination of hemolytic reactions because of the glucose content.

Kanamycin Aesculin Azide Agar is a selective isolation and enumeration medium for enterococci (Lancefield group D streptococci) in food, and is not intended for use in the diagnosis of disease or other conditions in humans. Sodium azide and kanamycin provide the selective inhibition required whilst aesculin and iron salts form an indicator system for the presumptive identification of enterococci. Incubation at 42°C will increase the medium’s selectivity.

Brain Heart Infusion Broth is used for the cultivation of a wide variety of fastidious organisms and is not intended for use in the diagnosis of disease or other conditions in humans. A rich isotonic infusion medium with tryptose (a mixture of meat and milk peptones) providing a wide range of substrates. A low concentration of glucose is used to stimulate early growth. The medium is lightly buffered to prevent the early death of some species due to acid production. Organisms which produce significant amounts of acid may well overwhelm the buffering system and auto-sterilize. The medium is suitable for use as a blood culture medium or as an enrichment broth for fastidious organisms.

MacConkey Agar No.2 is a modification of MacConkey Agar which contains bile salts No. 2 for the recognition of Enterococci, and is not intended for use in the diagnosis of disease or other conditions in humans. This agar especially useful when looking for Enterococci in the presence of coliforms and non-lactose fermenters from water, sewage and food products. Enterococci are frequently sought as an index of fecal pollution and appear on this medium as small, intensely colored red-purple colonies. Non-lactose fermenters appear colorless, whilst bile tolerant Gram-positive organisms, such as staphylococci and non-fecal streptococci, are completely inhibited.

m-Enterococcus Agar is used for the selective isolation and enumeration of enterococci by membrane filtration in a laboratory setting. m-Enterococcus Agar is not intended for use in the diagnosis of disease or other conditions in humans.
m-Enterococcus Agar was first described by Slanetz et al. for the enumeration of enterococci by the membrane filtration technique. In 1957, Slanetz and Bartley modified this medium by adding triphenyl tetrazolium chloride (TTC). Increased recovery and larger colonies were obtained by incubating the inoculated membranes on the agar surface instead of on pads saturated with liquid medium. The membrane filtration method is simple to perform, does not require confirmation, and permits a direct count of enterococci in 48 hours. m-Enterococcus Agar is also referred to as m-Azide Agar.
The presence of enterococci is a valuable bacterial indicator for determining the extend of fecal contamination of recreational surface waters. m-Enterococcus Agar is recommended for the detection of fecal streptococci using the membrane filtration technique for water testing. The food industry also has applications for testing enterococci using m-Enterococcus Agar.

Slanetz and Bartley Medium is used for the enumeration of enterococci from water samples as described in ISO 7899-2, and is not intended for use in the diagnosis of disease or other conditions in humans.
This medium was originally described by Slanetz and Bartley for the enumeration of enterococci from water samples using a membrane filtration technique, but it may also be used as a spread plate for the examination of other sample types. Enterococci reduce tetrazolium chloride to the insoluble red dye formazan, producing colonies which are dark red or maroon on the surface of the membrane or agar. This reaction is not exclusive to enterococci, and the count at this stage should be considered presumptive. Colonies may be confirmed as enterococci by demonstrating aesculin hydrolysis using Kanamycin Aesculin Azide Agar (NCM0198).
